Named for Pembroke W. Williams, a leading citizen of Bryan County during the 1890s, the city was the leading business and railroad center for the area in the early 20th century. Pembroke became the county seat in 1937 due to a County Referendum moving the county seat from Clyde (a town which no longer exists) to Pembroke, due to Pembroke's banking, business and transportation prominence.

Pembroke was home to the late Jack W. Shuman, a prominent state legislator and businessman in southeast Georgia during the 1960s and 1970s.
